FOXO4-DRI — the senolytic peptide.
FOXO4-DRI is a D-retro-inverso peptide designed to disrupt the interaction between FOXO4 and p53 in senescent cells, triggering their selective apoptosis. It is one of the most expensive research peptides on PeptideCompare, and one of the most scientifically interesting in longevity research.
How FOXO4-DRI targets senescent cells
Senescent cells — "zombie cells" that have stopped dividing but refuse to die — accumulate with age and secrete pro-inflammatory signals (SASP). In healthy cells, FOXO4 and p53 interact to maintain survival. FOXO4-DRI is a peptide that disrupts this interaction specifically in senescent cells, where this survival pathway is abnormally active, triggering apoptosis selectively. Normal cells are largely unaffected.
The original Nature Medicine paper (2017, van Deursen group) showed dramatic rejuvenation effects in naturally aged and chemotherapy-induced senescence mouse models — improved fitness, fur density, and renal function. This generated enormous research interest.
